Changing supply voltage from 5V to 3.3V will degrade performance and accuracy for both MCP3421 (INL vs. Supply Voltage) and AD8628 (Signal Voltage Gain).

The current source problem arises from using the 5V output line from the Arduino Nano.
As he wrote somewhere he couldn't figure out why the voltage (5V) dropped when the tips were...

That's what i described you would most likely find onboard a VFD.
A C scanning (multiplexing) the vfd (most likely thru an IC because voltage levels) and a serial (often 3 wire) interface...

As i recall they used a multiplex scan circuitry IC (maxim i believe) connect to a C. You might get better luck disassembling it and walking your way thru.

Test leads are unconnected (Needle in rest position):

VOM: ∞Ω -> Correct
VTCOM: 0Ω -> Incorrect
DMM: 'OL' = ∞Ω -> Correct

How come...? Take an example where the test leads are unconnected: both VOMs and DMMs will read 0 in V & A and 'infinite' or 'OL' in Ohm > Correct.
Unlike Vacuum Tube VOMs (VTVOMs) that will read 0...

Since the programmer, CH341A, is recognized both by OS and the software the CH341A isn't dead.
Because you've done the 3.3V voltage supply tweak if any input/output pins came in touch with 5V most...

Pins 6, 7, 8 are input pins and left floating in CH341A programmer.
See the attached schematic and try to figure out what's wrong (though there are several versions available).
